I have fresh dried blood spots that i need to extract Plasmodium falciparum parasite DNA for analysis of Antimalarial drugs Resistance markers.I will sequence the DNA thus i required high quality DNA.How do i go about the very first step ?

Buffer ATL is just a cell lysis buffer, you can google a recipe for one as there are plenty of protocols for different cell types.
It is important to have buffer AL though, as it contains guanidine which binds the the DNA to the silica membrane of the spin column.

Some of the modern mutated polymerases like KOD polymerase can amplify DNA from lysed blood so you might try just soaking in TE and amplifying direct and as your DNA amount will be very small try a nested pcr to re amplify a dilution of your first amplification
