You can dissolve around DNA to a concentration of around 10mM, but it is difficult to get that much to actually dissolve.
I'd try one of these strategies. Firstly, you can try adding the DNA very slowly to an aggressively stirred beaker over a long period (say a whole work day). Alternatively, you could place the beaker of water/DNA in a sonic bath and dissolve it that way, being careful that you don't allow the temperature to rise too much.
Hello Nasrin, DNA is acidic and in acidic environment does not dissolve (that's why you acidify with NaOAc pH 5.5 to precipitate it). If this conc. can dissolve (I never tried it, I think the max. I had was 20 mg/ml), try to do it in 10 mM Tris/HCl pH 7.4 or even 8 (basic pH is denaturing for the DNA, but 8 is low enough, will not damage anything). 10 mM is quite low, so it will not affect your future reactions that are normally performed at 50 mM buffer...
Warming up to 37 °C can also help, at least it will make it quicker. As well as shaking, or manually mixing every minute or so...
hi, well it will also depend on which type of DNA you are working with.Lets say if it is genomic DNA then stirring is not a good option since it can shear the high molecular weight DNA. If am asked to make a highly conc solution I would probably make a lower conc solution and then reduce the volume either by heating or filtration. I would also try different ionic strength buffers as well.
Use double distilled water (volume that you need) , make sure you don't dissolve in very less . Keep the DNA sample at 60 degree dry bath for about an hour. I usually do this but i do not know the exact concentration that i took. so you can try this taking less amount of DNA so that your stock is not lost. OR try with TE buffer.
You should try to dissolve your DNA in T.E buffer .the composition of this buffer is 10 mM Tris of PH 8 and 1mM EDTA of PH 8 . you can leave your DNA overnight at 37C well you should not dissolve it all together make aliquots of your DNA .
Or you can store your DNA for years while making many aliqouts with the salt and 95% ethanol and store it at -150 degree freezer and precipitate it before using instead of dissolving all of your DNA.
