I want to graft the cancer cells i.e, MCF7 on cam of egg. To do this I need to have some scaffold. In a paper i read about matrigel, is there any other alternative of matrigel or can we graft the cells directly onto the cam without any scaffold?
You can make aggregates of your cells using a silane-treated flask (to prevent sticking) and rotary aggregation. Gentle centrifugation will give you a mass of aggregates that can engraft on the CAM. The CAM may grow around your graft, so harvest any lumps of membrane. Remember that the immune system of the host embryo turns after day 17, so harvest before that.
