I am planning to send few samples for RNA-seq. I have always used RNAeasy kit from Qiagen to extract mRNA and I am really comfortable with it. But I ran into some conflicting suggestions with someone who did RNA-seq that TRIzol extraction was used in their approach.
Any suggestions or insights as to which one is better?
Dear Michael, the RNA extraction method is not an issue as long as the RNA quality is good. Unless you are working on a very difficult sample, the quantity is not a problem, even though different extraction protocols can lead to remarkably different results in terms of extraction yield. Sequencing centres usually require a very low amount of total RNA for the library preparation (250 nanograms to 1 microgram, depending on the service provider).
Quality is a much more important parameter and my suggestion is to choose the most appropriate kit based on the best absorbance ratios you can get from your sample.
