I stored them by glycerol 40% which equally added to LB broth. They had function after 2 months but now after 4 months they don't have ability to produce plaque on plate.
Hi Reza. I process and store my phages as follows: (1) After ultra-centrifugation, I collect the phage band and add an equal volume of sterile phage buffer (sodium chloride, magnesium sulfate, Tris-HCl, gelatin, water) and store this at 4deg; (2) I pellet the phage band also through centrifugation, resuspend this pellet in phage buffer at 1/10 original volume of band and store at 4deg.
I have screened several different storage temperatures, durations and storage reagents. These two methods have provided the best results. Phages showed virulence even after 18mo of storage. Admittedly, titers did drop but at its highest, titers had decreased by 20% of the original. I do hope that this helps! Iona
Hi Reza. I store my phages adding 40% glycerol too, but I prepare this solution in lambda buffer (60% lambda buffer + 40% glycerol), and keep them at 4C. Freezing and thawing lower the titer. You may want to check your host, frequently the phage is OK but the host is the one that loses its ability to support phage amplification.
