I isolated phage against Pseudomanas aeruginosa PAO from sewage. There is a need to have sufficient pellet for SDS-PAGE. I recruited PEG 8000 (40% PEG+ 0.5M NaCl ) in equal volume for precipitation. 10 cc of phage mixed by 10 cc of PEG and incubate at 4 c for 2 hrs. Then centrifuged for 15 min at 12000 rpm.
Always there is not any things but sometimes there is a clear and pale pellet. When I used this pellet for SDS-PAGE there is not any band while ladder completely separated and host bacteria has obvious bands.
The last image down below seems to be of Poaceae. The other two pollens are difficult to identify with the present LM image. A distinct exine structure and surface image of the pollen is needed to identify the pollen type. I suggest you to correlate it with the kind of plant species available in the vicinity of the sampling site (within 1km radius). Prepare pollen reference slide of all the flowering plant species available in your area will help you in identifying these pollens.
We have done SDS-PAGE for bacteriophage against B. abortus there for concentration of eluted phage we used Ultacentifugation at 200000g at 4 c for 4 hours in ultracentrifuge (Hitachi, Japan). You can try this. Here I am attaching a link, they have done SDS-PAGE on phage against P.aeruginosa PAO.
