Hi all,

I need to do a western blot of a protein where one isoform is ER-membrane-bound. My antibodies do not show that isoform reliably and someone suggested it could be lost due to my RIPA buffer based isolation. The cells I'm working with are iPSC-derived cortical neurons.

Does any of you use a different isolation protocol to preserve the membrane-bound proteins?

Or does anyone know a subcellular fractionation protocol suitable for neuronal cell culture?

How many cells do you commonly use to obtain enough protein there?
