Dear colleagues, I want to know how can I prepare the Giemsa stain as the last time I prepared it , it was light, not stable, and be faint after 2 days

Since you unfortunately have NOT specified what purpose the GIEMSA staining solution should serve it might be essential and of benefit if you let readers know what you are going to stain with....(There are several "Giemsa-stains" and "Giemsa-Modifications", so the more accurate the details of your task, the more readers' answers will be on the spot...) I do not know exactly what you are going to stain, but the quality of the dye (GIEMSA) and the solvents are important. The quality depends on the manufacturer. It is also important stores the Giemsa solution in the amber botlle, avoidind direct expositure to light. Good luck, Ida
NB: Naturally quality of the dye (and other chemicals to be used), its/their source/manufacturer, storage mode and staining conditions are important parameters for successful staining. As for the quality of stain/dye one should read about the certification of dye(s) (e. g., by the ).
