Immunoblot bands of protein size below 20KDa (19, 15, 12) becomes curved, I use PC9 cell. However bands for larger protein size has no problem. I use 12% and 15% SDS Gel and gel running at 100 volts and transfer at 90 volts for 2 hours. What change should I do to get nice and beautiful bands?
Simplest solution is to use pre-cast gel...!!!!!! otherwise make sure you take plenty of time to cast resolving gel may be a day advance.....but once you cast the stacking gel, you should not wait for very long to load your samples and start the run... once the dye shows a clear stacking at the top of resolving gel and moved in the resolving gel then you can increase the volts from 100 to 120. Bands get curved like "smiling" or "frowning" depending on many issues some of them can be using old buffers, poor casting of gel and last but not least problem with the loading dye... try to use everything fresh and hope these suggestions help....
