For my plasmid isolation the protocol says to use 2 M acetic acid but our store has only got 100 % pure glacial acetic acid. So how can I make a 2 M acetic acid? Happy for any help!
Glacial acetic acid is 17.4M. From that, you can use the M1V1=M2V2 (where M=molarity and V=volume) equation to calculate the volume of glacial acetic acid you'd need for however much 2M you're making. So:
17.4M x (Volume of glacial acetic acid needed) = 2M x (Volume of 2M acid you need)
We suppose that you are going to make 25 ml acetic acid 2M.
m1v1=m2v2
2×25=17.48v2=> v2=2.86 ml
Dilute 2.86 ml of 100% pure glacial acid to the total amount of 25 ml in a volumetric flask with deionized water. (or simply add 22.14 ml of deionized water to 2.86 ml of acetic acid)
@Yemisi Omonije: Assay is nothing but purity in percentage and usually it is labeled in chemicals nameplate or you can get it in the manufactures product webpage.
