I differentiate N2a cells with 20 uM retinoic acid 1% serum in a 24 well plate. During the differentiation process that lasts 4 days, I change the medium every two days, but on the 4th day the cells start to die. How can i make cells live for a long time (10 days)?
Are there any specific mutations on the line you’re using? How have they’ve been cultured before. When it comes to neuronal cells, substrate plays a big rule- Try to find out the conditions that line has been subjected to before you took over.
Matrigel, polyornithine and Neurotrophic cell media are always good choices to consider

Hello Mani Garg, i tried different concentrations of retinoic acid and serum, but I did not perform well. There were suggestions for me to use different media, but I couldn't try them because I didn't have them.
