I'd like to characterize an unknown bacteria strain which was stored in a glycerol stock at -80 degrees Celcius. I've inoculated the unknown bacteria strain on an LB agar plate. The colonies of this bacteria strain were inoculated in a flask with 10 mL LB medium shaking at 200 rpm and 37 degrees Celcius. I want to measure an OD600 of 0,1 at least, but the bacteria strain barely grows after incubating for 24 hours. The bacteria strain doesn't grow in Tryptone Soya Broth either.
Does preheating the flask with LB medium increase the OD600 before I inoculate it with the bacteria strain?
