I am trying to revive Pseudomonas aeruginosa from frozen glycerol stock and also from cryobeads. But there was no bacterial growth. I used nutrient agar, tryptone soy agar to streak on. Also I added glycerol stock to nutrient broth and tryptone soy broth but there was no growth.
I would appreciate your guidance.

Pseudomonas aeruginosa had been considered as an obligately aerobic bacterium previously, but it is now recognized to be highly adapted to anaerobic conditions.

It's been considered facultative throughout my 40+ year. On can find occasional isolated descriptions as aerobic and I doubt the original description in German included relevant info. It's described as facultative in my 1957 Bergey's and understanding of the facultative nature of some Pseudomonas spp. goes back > 100 years.
A colleague used high sucrose and yeast extract broth medium to revive fungal cultures. High sucrose would prob be inhibitory but maybe try NA/NB with added glucose and 1% yeast extract. You might add some activated charcoal - some folks think it eliminates trace inhibitory material like fatty acids.
In any case, you need to find a better way to preserve your isolates.
Phil Geis does it work for Pseudomonas as well? I do have carbon and nitrogen supplements in lab. Can I try with that?
