I'm working on freshwater nematodes for my undergraduate thesis.
After I fish them from sediment or water under the stereomicroscope, do I have to place them on a temporary mount and ID them on the compound microscope? Or should I fix them on a permanent mount instead?
I've been having a difficult time identifying the nematodes, especially those which are highly motile.

First of all kill and fix them by pouring boiling hot 3-4% formlin or 70% alcohol. After that clear them in lactophenol.
If your purpose is just to identify you can prepare a temporary mount. Take a drop of water on a glass slide and transfer the nematodes. If the nemas move actively just warm up the slide. Now, settle the nemas on the surface of the slide. Cover with a cover slip. Observe then

I second the answer by Mohammad Mahamood. The bodies of live nematodes will stay intact, which you need for ID. If they're moving around too rapidly to get a clear view, you can place them on a hot plate for just a few moments (not enough to evaporate the water!). You can also seal the cover slip to the slide with clear nail polish around the edges.
We all know that nematodes are very much transparent. If you want to identify them, you need to fix them with hot fixative and then make a permanent slides of them and only then you can identify them easily.
