Can anyone send me a reference on how to extract compounds from bacterial agar plate, as the particular bacteria shows the phenomenon only on agar plate.
Try to add to the surface of the agar medium a buffer phosphate saline at pH7 and leave 24 h at 4 C and then take the buffer with a pippete to a centrifuge tube and concentrate the compound by centrifugation. If not try to use a soft agar layer cover.
It is really astonishing that your bacteria is able to show the activity or produce a compound only on agar plate because Agar Agar is merely a solidifying agent. I would suggest to again grow the bacteria in the liquid media and test the presence of compound in the supertant if it is secreted extracellularly. If you do not detect any compound in the supernatant it means the said compound is intracellular and in that case you need to disrupt the cells and extract the compound. Even if you do not get any activity then you may follow the method as described by Dr. Hussein. Good luck to you for your research.
For enzymatic activities i freeze the agar, after removing the colony, and frozen i centrifuge at maximum speed. Then i used the exudate for further analysis.
