We have been using electroporation to transfect a hybridoma cell line with siRNA and got knockdown efficiencies between 50 and 70% which apparently is quite good for that method and that cell type. Nevertheless, I would need something more efficient and ideally in a human antibody-secreting plasmacytoma cell line. Can anyone help? Screening the literature it seems that this is not the most straightforward task... Does anyone have experience with e.g. lentiviral transfection or Crispr-Cas9?
