If you consumed the entire sample - try inoc'ing 1490 with the some of the thio broth (you prob already tried that)

I understand that you're facing an issue with reconstituting B. ovatus ATCC. Despite placing it in THIO broth, you didn't observe any turbidity after 24 hours. This could be due to several reasons:

Possible Causes:

Inadequate rehydration: The bacterium might not have been properly rehydrated from the freeze-dried state.

Incorrect storage: The freeze-dried bacterium might have been exposed to temperature fluctuations, moisture, or light, leading to viability loss.

Contamination: The broth or equipment might be contaminated, preventing the growth of B. ovatus.

Old or inactive culture: The B. ovatus ATCC culture might be old or inactive, leading to reduced viability or no growth.

Suggestions:

Check the rehydration process: Ensure that the bacterium was properly rehydrated according to the manufacturer's instructions.

Verify storage conditions: Confirm that the freeze-dried bacterium was stored correctly, following the recommended storage conditions.

Sterilize equipment and broth: Ensure that all equipment and broth are sterilized to prevent contamination.

Try a fresh culture: If possible, obtain a fresh culture of B. ovatus ATCC to rule out any issues with the current culture.

Incubate for a longer period: Incubate the broth for a longer period (e.g., 48-72 hours) to allow for slower-growing bacteria.
