There are many theories trying to explain ways to create a stable transfection in mammalian cells, by selection. Does it work? Is there any other method?
Hi Panagiotis...the pCMVTNT plasmid does not have an eucariotic antibiotic resistance, neither is viral...so you cannot use it for stable transfection..(or maybe you can but the method is more cumbersome and not secure than normal generation of stable cells). I suggest you to clone in another vector that has the selection marker....and unless you do not want to start playing with Cas9/Crispr system, selection is the only system to obtain a stable overexpressing line...good luck!
