I am isolating Actinobacteria from plant rhizosphere on Agar medium. But i feel tough in differentiate Actinobacteria from others bacteria on agar medium. Can anyone help me what are characteristics of Actinobacteria on agar medium?
I used air dried method as pretreatment before i made serial dilution. and i am using Mannitol soy agar & Tap water yeast extract agar as isolation media.
but i am still confused, which microbes that I should purificate, Sir hehe
In general, the actinomycetes colonies on agar plates are characterized by:
- It is not easy to pick them up by the loop because they embedded into the agar except the rough surface (In most species). On Contrary, the colonies of fungi and bacteria are so easy to pick them up by the loop. In addition, the fungal mycelium is long and visible by naked eye.
- Actinomycetes form not spread small colonies of chalk touch.
Actinomycetes have chalky texture. They have aerial mycelium with different color. Also, they have background with different color. Sometimes there are colored pigments in the agar.
