I am experimenting with the plant extract containing high amount of decursin in in vitro cytotoxicity study. This compound is soluble in DMSO / 100% ethanol but completely insoluble in water. The prescribed dose for my study is (approximately 0 -10 mg), so when I apply DMSO solution of that compound to my cell culture media (obviously water based) at the desired dose, it forms a precipitate and makes the culture medium turbid. It will be very helpful if anybody can suggest a way out of this problem so that I can use this compound for my system.

I agree Chibuike's points to some extend, but still there could be a precipitation occur. The solubility of this compound depends on the final content of hydrophobic solvent/DMSO in the whole mixture. If you add little volume of highly concentrated drug in DMSO to the medium, it is still the same amount drug you want to dissolve in mainly water, which will not work then.
The "dosage" you describe should be a concentration, not a mass. Try to dissolve the drug in water alone (use DMSO stock), do different concentrations. With that, you'll know the max. concentration you can go for the medium without precipitation. "Completely insoluble" as you describe is not the right term, it will be soluable to some extend. This is necessary for well working drugs - if they are too hydrophilic, they'll get externalized from the kidneys very quick and can not have any positive effect of curing the disease. If they are too hydrophobic, they'll accumulate in the body and cause further problems. The balance is important (Doxorubicin for instance).
The effect of the drug treatment also depends on the time the drug is in contact with the cells.

If your extract dissolves well in methanol, I will suggest to prepare a highly concentrated stock solution in methanol (in some cases, sonication for few minutes may help). Then add small volume to the cell culture (so the maximum methanol concentration in the medium does not exceed 0.1%). GOOD LUCK
hi, i have faced the same problem recently, what i did was....... suppose i want to make 1mg/ml concentration, i dissolved 10 mg of my extract in 1 ml of pure dmso and add 9 ml of mem and filtered with 0.2micron syringe filter... i used this as a stock solution and made further dilution with mem as desired ... if u dissolve ur extract in pure dmso, it will precipitate again when u add ur extract to ur culture..... all the best
