I am trying to develop MCF-7 breast cancer cells resistant to doxorubicin , I reached 60 nanomolar concentration, the cells are getting resistant but they are not growing anymore , I need to grow them because I need enough cells to complete my experiments, whenever I change the media on the cells i add 60nM dose with it , I can not add media without the drug so that the cells do not lose the resistance character they acquired, is there any clue how I can increase their growth without losing resistance-character? Could anyone thankfully help . I have attached a photo for them.
Try growing them without Doxorubicin next time. I dont think they will loose their resistance.
When the cells acquire resistance they will not loose it. Remove the doxorubicin grow them to have enough cells and do the experiment. Remember, You might be selecting resistant clones in your cell line rather than generating resistant cells.
