Triton X-100 has been widely used to permeablize cells for flow cytometry. How can I choose the concentration of Triton X-100 if I would like to detect expression of one protein in the whole cell? And would the concentration influence the results? For example, there would be differences between 1% Triton and 0.1% Triton?

Hello, did you by any chance find an answer for your question? We are used to do membrane markers for FACS in our lab and we are now considering intracellular marker but we haven't found a protocol using formaldehyde fixation and permeabilization that does not require a kit.
