I have tried everything, high purity sterile media, adding fungizone, sterilizing the flow devices, the incubator, the incubator water, but I still can't shake off the fungal infection I get only 24 hours after I start the flow experiments on primary cell culture. is it possible the fungus is resistant to the fungisone I use? do you recommend a broad spectrum anti-fungal?
its possible that the cell that you use infect with fungi and the fungisone is not effect on fungal,
Fungal spore contamination is one of the toughest to get rid off. It is a strong possibility that your experimental instruments and hardware is infested with spores. Try to culture in a differant lab setup to confirm this

we had a similar persistent problem just a few weeks ago, where all cleaning and renewing of solutions and cell stocks did not help. We noted that the problems mostly occured when someone else was working in parallel in the same room (which should ideally not be the case). Finally, we could pinpoint it to a problem with the laminar flow hood itself (weak and fuzzy airflow). When was the last technical check on the hood? Ideally, it should be tested once a year. Also remember that older models have much higher thresholds regarding the air flow than modern ones, so the faulty device may even pass the test although the flow is weak or irregular (as it was the case with our hood).
Check your serum sample, contaminated serum samples would continue fungal growth. Contamination of cells is also possible. Wash the cells two to three times with double the con of fungizides.in pbs. if not work start with a new cell line.

Try to use Amphotericin-B and problem seems with u might be an indeginous contamination in your tissue sample which gives u false impression of contam due to your culture settings.
First try to make sure that tissue u use is sterile (I hope it's not a skin sample). Second, try to give extensive washngs to ur tissue with PBS even after you chop it down. Repeated washings are an easy survival tactic in case of primary culture. Third, Change the media next day immediately in morning once u seed the primary culture as it will help to eradicate non adhered cells/tissues and some slow tissue contaminants
Once u get sure of it other precautionary measures are sufficient to prevent any fungal contam.

Have u decontaminated the hood and ideally changed the filters? All cleaning is worthless in case you have spores in the hood
