I'm using a protocol proposed by Bailey et al. 1992 to measure xylanase activity from a liquid form of enzymes in this protocol I use a dose = 0.1 ml of enzyme . I'm wondering if I can use the same protocol to measure xylanase activity from a powder form enzyme or from a probiotic.

Dissolve the powder gently in a suitable buffer, taking note of the concentration in mg/ml, or U/ml if the powder is supplied with U/mg information.
Thanks Sir for your answer. But sometimes my enzyme product is insoluble. Should I use it in solution with buffer even it does not dissolved?
Undissolved enzyme will probably not be active. Either dilute it more until it is dissolved, or try other buffer conditions in which it is more soluble. If this is a commercial product, ask the supplier how to dissolve it.
