House keeping gene VS cDNA concentration.

Dear Colleagues and Mentors,

Which one is better to follow to measure the expression of gene? Keeping one/set of house keeping gene(s) or direct measure of cDNA concentrations to normalize the raw data, which one is better or more convenient? Seeking your suggestions.

The quality of normalized quantitative expression data cannot be better than the quality of the normalizer itself. Optimum number of housekeeping gens should be nomalized for RT-PCR analysis. To identify the most suitable housekeeping genes, mRNA levels will be quantfied using cDNA concentration, then you can proceed further with idenfication of suitable reference genes.
