We are trying to set one up in our laboratory and we prefer an actual lab protocol and not one that's kit-based. Would anyone know of a Haemophilus influenzae type b ELISA protocol where buffers are prepared from scratch and we just obtain reference standards and QCs from other institutions or, biomedical companies?
Buffers should be standard do you have antibodies for HiB or are you checking serums or other tissue samples?
Goolgle ELISA protocols (add bacteria if you are detecting a whole bacteria you may need poly-lysine) you will get prescription for all buffers need.
Thanks Ehud! Yes, we're checking sera and plan to use a Hib capsular polysaccharide conjugated to human serum albumin for detection.
There sould be no problem binding directly to ELISA plates or even do sandwich with anti HSA as capture antibodies.
