I am analyzing the antibacterial effect of my postbiotic on Streptococcus mutans as a pathogen. I found different methods with some variations; some suggest an inoculation volume of 10 or 20 microliters, while others recommend inoculating 100 microliters of bacteria into each well containing 100 microliters of the drug. If anyone has experience with this test or working with this strain, I would appreciate it if you could share your insights with me.
There is an standard protocol for MIC.
I'm suggesting an article please go through it and you have to decide the range of concentrations that you want to have for your postbiotic testing.

To study MIC, any of the methods established by EUCAST or CLSI for streptococci could be used. If you want to know the MBC as well, the most appropriate method would be to use a broth dilution method, in order to establish not only the inhibition percentages, but also the bacterial death percentages.
