I want to co-culture T cell and tumor cell to see T cell cytokines release. I used to co-culture them and add Golgiplug immediately for 6 hours. But now the cytokine signal is weak. Is it helpful to co-culture them more than 6 hours? Can I co-culture them without Golgiplug for maybe 12, 24 hours, and then add Golgiplug to keep cytokine in cells? Thank you
Hi, you should add the Golgiplug 1h after the stimulation (to block the cytokine secretion).
Hi Maud, Thank you for your answer. Do you have experience to add Golgiplug immediately when you co-culture effector and target cells? Is there remarkable difference? how many hours do you usually keep golgiplug in the media to show sufficient cytokine release intensity. I keep Golgiplug in media usually 5-6 hours, but cytokine intensity is not high enough. So I am wondering if co-culture with Golgiplug more time like 9 hours. But according to protocol, keep Golgiplug in media no longer than 12 hours.
