Yes we do it regularly. It depends on the particular gene. If it is regulated in the transcriptional level, the gene expression and western blot data will show a similar up/down regulation pattern.
I confirmed all my real-time PCR data with western blots. Though it must be emphasized that changes in gene expression don't necessarily mean that there is a change at the protein level. This must never be assumed!
Thanks Alice. 0.6 for all genes is qctually somewhat high . i was expecting lower level in a way. but i think this might go up dependinf on the biological function and abundance of protein.
Depends. Some genes are mainly regulated at the protein level, by changes in the degradation rate. Translation rates of RNA also vary, and could be regulated.
