I have grown Adhaeribacter aerophilus in R2A and R2A broth . It grew well on R2A but did not grow on R2A broth at 30 degree C . Today is 7th day of incubation but result is same. I am doing this to preserve the bacteria at -70 degree.
Why can't you save your regular growth at -70C? Just spin your culture down, resuspend it in 1 mL of nutrient broth +50 % glycerol and put it away at -70.
Description of the species reports growth on R2A agar beyond 30C - http://ijs.microbiologyresearch.org/content/journal/ijsem/10.1099/ijs.0.018374-0#tab2. They reported growth in R2A broth across a pH range but did not mention temp of incubation. Have you tried a lower temp of incubation?
Actually I need to make stock for A. aero preservation . Usually our lab culture bacteria in broth and incubate it in 30 degree shaking incubator after two days bacteria grows and put 1ml of it into eptube , spin and culture down and send it for sequencing but I found nothing because the bacteria was not grown on broth
If you haven't succeeded to grow your strain in broth try to prepare agar water because the agar might contain some special salts etc. supporting growth of your strain. Agar water is easily prepared. Suspend 15 g of agar agar in 1 l of water, let the agar settle down (can be accelerated by centrifugation) and use the supernatant for preparing the broth.
