I am looking for simple way to detect human tissues implanted in athymic rodent. If anyone has used the anti-nuclei antibody or other antibody, please let me know how it worked?

I have used this antibody and it works really well. Used 1:200 concentration on cryosections of mouse retinas transplanted with human cells. Its a mouse antibody so be aware of the background on your tissue. The conjugated version of this antibody also works well and decreases background.

Yes, it works well in cryosections without any PFA fixation.
It works on frozen sections only at a much higher concentration of 1:20.

It works on both paraffin and cryosections for both mouse and rat tissues to detect human cells. In our lab we have used 1:100 dilution for cryosection and 1:75 for paraffin embedded sections. Also please check exact protein concentration of the antibody and confirm the staining using exact concentration corresponding isotype antibody.

Is this IHC assay alone of localization of human cells in rat or mouse tissues is well accepted for tracing of human cells?
Do we need to do any other assay for further support. Since some paper has done ISH by Alu sequences etc.
We used it for cryosections (1:100) and it worked, however we could not get any signal for paraffin samples.
