I am trying to reprogram a mouse liver cancer cell line to iPSC. About 2 weeks after transfection I started seeing mouse ES-like colonies. I dissociated and replated them on new feeders hoping to get more colonies, but what I saw were not colonies, rather cells with morphology similar to the original cancer cells. May I ask what those colonies might be?
EMT and you can study some modifications on the molecular pathways .
It can be done with some cancer types, for example MLL-AF9 leukemia, which is largely driven by epigenetic aberration.

However, some driver mutations/rearrangements may prevent the iPSC from being as malleable as other non-cancerous cells in terms of reprogramming potential.
I think it's not a real colonies. The cells are not fully reprogrammed. This is not a successful experiment

Andrew: Is there a way to know whether the cancer cell types I am trying are not reprogrammable? Or if I can't after much trying then I will know?
Yanfeng: May I ask, how should the cells look like when they are on the right track to being reprogrammed? There are loads of cell clusters of various morphologies in my cultures. Any tips what to look for?
