it is depend on kind of your cell target, in my experience, I isolate the mesenchymal stem cell from adipose tissue from liposuction with mechanical and collagenase.
First of all you should loose the cell-ECM connections (integrins). Use EDTA or EGTA as Calcium chelators . For a large piece of an organ, perfusion is a good idea.
Then you can use enzymatic digestion and after neutralizing the enzyme, isolate single cells.
I agree that enzymatic digestion is the best way to go. If you have access to a cell sorter (or a core facility with a flow sorter), good sorters can deposit a single cell into individual wells.
