I want to study if bacterias are associated with root, can anyone suggest for fluorescent marker which would be better to use? or which method is good (example some staining solution like acridine orange etc.) ?
You may use Fluorescent staining using CTC or membrane permeable fluorescent dyes like SYT09 and DAPI. The following combination works well: 5 micro mole SYT09, 30 micro mole propidium iodine and 0.1% saponin. For fluorescent staining of bacterial endospores please see the attached files.
in my opinion for identification of the bacteria we should tag the bacteria with gfp marker which gives green fluorescent light under UV rays. thus we can identify the marked strain very easily. we can also use fluorescent dye and observe under flourescent microscope. another method is the ELISSA test for easy identification of bacteria.
