Its taking me ages to optimize the ELISA to measure cortisol in sheep (ovies aries) plasma, and its also too expensive if I'm going for an commercially available ELISA kit as the sample numbers I've got with me is too big!!
Inputs on this are much appreciated !!

If you want to use niether Kits nor commercial antibodies, you have any difficulties during your experiment and setting it up.
In my country, Iran, these kits are not too expensive and using of them is routin.

thanks for looking into my query.
I'm following the Sandiwch ELISA protocol in general for my cortisol experiments, and same with RIA for testosterone. But, the issue is with ELISA, as I've now managed to sort out the RIA protocol. I'm using the commercial available antibodies for ELISA starting from coating till adding the HRP conjugate, however, every time i conduct the experiment, it doesn't work the every second time which is depicted through the standard curve, because of which I'm scared of loosing my plasma samples.
I now have understood that validating ELISA takes ages and that's not what I should be doing in my PhD (give me the experience though) unlike how they do in the companies.
When it comes to ELISA kits for cortisol, they are expensive in UK,however, I'm not sure whether we can import it from your country or it might end up costing the same after all the import duties being paid.
