For THP-1 differentiation we use reconstituted PMA at 1mg/ml.
Our final concentration added to cells is 50ng/ml.
Since final working concentration & volume required is low, we usually make an intermediate solution. Intermediate solution is made at 100ug/ml or 1ug/ml.
Does anyone know how long we can store the intermediate solution?
Can we reuse it after storing at -20 C?

You make a stock solution of PMA in DMSO and aliquot the stock. Such a stock solution may be used up to about 3 months if stored at 4 °C in the dark. PMA prepared in DMSO solution stored at –20 °C in the dark show no degradation of PMA for at least 6 months.
You do have to make intermediate solutions (in aqueous medium) and it is not recommended to store these solutions. You will have to discard them every time you make such solutions as they are not stable. Each time you will have to make fresh dilutions in aqueous medium. You cannot reuse the intermediate diluted solutions except on the same day if you are going to perform another experiment.
