I am using Minimal SD base + DO amino acid mixes to make SC agar plates for my yeast chemical transformations. This minimal SD base contains glucose, though the directions say to autoclave at 121C for 15 minutes. The color does change slightly more yellow after autoclaving, indicating some caramelization. Should I add more glucose to this media after autoclaving? I always assumed that the slight caramelization isn't a huge problem because the plates only need 2% glucose so it shouldn't make a huge difference, but my transformation efficiencies have been extremely low, suggesting that there is something going on.

Note: As I am making agar plates, I cannot filter sterilize as I need the agar to melt.
