what ca we do if we are sure that one of 100 of bacteria has desired gene (which we have its DNA sequence) and we want to know which one has it? to explain more:
I have sequenced 100 fosmid with NGS in one pool, after NGS data analysis I have found my desired gene, as I have pooled all the fosmids together now I am not able to know the origin of my gene.
Is there any method to find out the origin of the gene?
Plate them out and PCR each clone. Or, for only 20 reactions: array them on a 96 well plate, pool each row and each column and PCR the pools. The positive column and positive row will give you the well.
