I plan to silence a hypothetical tumor suppressor gene in two cell lines. I was first convinced that a stable transfection with shRNA was best, although at the beginning it requires more work. Should I provide a better model for understanding the role of my gene? I think this is the best way particularly because my cells are not so efficiently transfected (let's say about 20%). However going through the literature I found many people transfect with siRNA (sometimes conjugated) and I start having some doubt if my choice is the correct.

Can you help me in clarifying which should be the best strategy?
