Plasma is required for miRNA analysis but in our settings we have to take sample from long distant place sometime from outskirts so that the sample(plasma) seperated cant be stored straight away to the freezer. In such condition if the plasma remains at room temperature of 25-30C for 24hrs before placing it at -80C will it be Ok for miRNA analysis??
It is good idea you place the sample in LN2 right away. Or at the max on dry ice right away. I am sure mRNA will be severely affected leaving at RT for so long. You can compare one set between 2 conditions.
Yo'd better use liquid nitrogen container for transportation of samples.
You could also sample the blood in a blood collection tube that has improved preservatives that stabilizes cfRNA and exosome counts for up to 7 days at room temperature. You might have a look at the Streck BCT tubes. https://www.streck.com/collection/rna-complete-bct/
Sample the blood at any distant location and ship the whole blood to your central facility to centrifuge and store your plasma.
