I want to detect calcium release from my protein in pH 4.5 aqueous buffer by dyes such as Calcium green, but I am not sure whether these will work at pH 4.5 and will it be soluble? Does anyone have information regarding other calcium detecting derivatives, which work under these (low pH) conditions?
It is a problem to detect calcium at pH4.5. Calcium green is soluble in such conditions, but has another affinity than at pH 7 and much lower quantum yield. So, you should found (or measure) stability constant of Calcium green complex at pH 4.5.
