I want to construct a calcium standard curve using Calcium-Green dye and my own 1-39 uM calcium chloride solution. Is it possible to construct a good standard curve without using the commercial Calibration kit which contains EGTA solutions?
The problem is that the assay is not working. All Calcium concentrations give similar fluorescence intensity. I will try lower concentrations (0- 1 uM).
I think it may not be possible to get a good standard curve of calcium without using K2-EGTA and Ca-EGTA mixtures. Even my controls without calcium gives very high fluorescence (I am using high quality milli-Q water). Can somebody suggest any other reagent to quantify calcium in a metalloprotease in 1-10 micromolar range. Any personal experience of using these kits will be very useful.
