Are there any well established fluorescent dyes (alternatively to PicoGreen) that can differentiate between supercoiled and relaxed DNA, so that they are able to measure the difference in supercoiling of plasmid?
Hello,when you load plasmid DNA on the gel, you see supercoiled DNA move faster than relaxe DNA. Infact Supercoiled DNA is like a small ball that can move faster.You see it in the below of gell lower than the other.good luck
We used to run an agarose gel without nucleic dyes and stain the gel with GelRed (Biotium). Then, a photo of the gel is taken and analyzed using ImageJ. The intensity of supercoiled and relaxed plasmid can be calculated by the ImageJ.
