I am trying to make cDNA from miRNA for qPCR. I am planning to go for kits. Most of them uses polyadenylation. I wonder if mRNA also gets adenylate with the kit protocol? also which kit is best for miRNA RT-q-PCR?
Yes, there may be the chances for mRNA polyadenylation, so you need to design miRNA specific primer which only amplify your target. The best way for miRNA profiling is stem loop qRT-PCR. As the RT-PCR is highly sensitive so you should design your primer more specific.
