I have a small molecule reagent with a boc-protected amino group. Now the reaction needs to be reacted at 37°C for 30 h. Is this protection group stable?
Dear Shuai Peng, I think if it is not in an acidic medium, protecting boc thermal stability is favored, because deprotection requires temperatures far above 37°C. Please have a look at the links below. My Regards
N-t-Butyloxycarbonyl group is progressively labile with the increase of the acidity (both, Lewis and Brønsted) of the reaction media. To keep the Boc protecting group intact, one should make sure that the reaction is carried out under basic conditions regardless of temperature of the medium.
