I need to express a 5 AA long peptide in any suitable mammalian expression vector. CI fear that if its allowed to be expressed in pCDNA3.1, there might be a leaky overhangs of extra amino acids coming from the downstream seq of the promoter ( even if we put extra start codon in-front of the peptide nucleotide sequence), which would compromise the activity of peptide. Could any please suggest me the literature regarding the precise expression of small peptides in any mammalian expression vectors?
Why do you need it to be expressed in a mammalian vector? Such short peptides are better to let chemically synthesized.
It is not clear why you want to express such a small peptide in mammalian cells? It is easy and cheaper to chemically synthesise the peptide rather than going through all sorts of issues in expressing in mammalian cells.
I have done my assays with the peptide. Expressing a peptide in a mammalian system is sort of a followed up assay, towards developing a FRET reporter construct.
