Moreover, after centrifugation, the pellet was not dispersed in pure deionized water even upon vortexing and sonicating. What should be the speed and time for centrifugation? Need Suggestions!
This Is advisable, the ultrasound treatment can decrease the PDI and size NPs. So, you must control the temperature if your drug can degrade. Moreover, the full caracterización (PDI, size, zeta potential AND encapsulation efficiency) is requiered.

Ultrasound creates powerful compression and stretching of the dispersed system. Depending on the applied power, even the decomposition of water into free radicals can occur by breaking the covalent bond. Covalent bonds in chitosan can also be broken. So the particles get smaller. We used the effect of ultrasound on the size of surfactant micelles.

Encapsulation, like the formation of micelles, is also regulated by hydrophobic interactions. Therefore, the destruction of your encapsulated structures is possible.

From my experience, ultrasonication for reducing the particle size or obtaining arrow size range is not safe, because in many situations the encapsulant leaked or the nanoparticles were broken-down. so I am recommend using homogenizer or screening the surfactant type/ percentage to reduce the surface tension of chitosan polymer that will reduce the size and PDI significantly
Chitosan nanoparticles can be of course sonicated if you want to get smaller particle size but don't raise the temperature too high as it may have deterioration effect on the polymer.
Hazem Choukaife which surfactant would you suggest? I'm using Tween-60 (up to 3% w/v) with encapsulant. But without an encapsulant, is it necessary to use any surfactant? Can chitosan NPs be prepared using merely chitosan & TPP?

Normally I use tween 80 (1-3% ), or try to use Span 80 in organic/oil phase. Also you can use both. As well as something so important is the M.W of the chitosan. If is it too high you cannot reach very tiny size.
