I am isolating the E. fungi from grasses for extraction of secondary metabolites and i should preserve the culture for further work, hence i am using liquid paraffin for the preservation but by using this mineral oil i should recover that fungal culture by three or four time sub-cultures by doing this the fungi may lose the metabolites. so can any one suggest me what can i follow..
1. Go for glycerol stock preservation: Grow the fungal culture in log phase, harvest biomass and then preserve it in 30% glycerol at -80. subculture once in 6 month
2. For long term storage (>1 years), try lyophilizing the cultures and seal under the vacuum.
Glycerol stock is a good option. you can pre aliquot your fungal stock in multiple 0.2 ml tubes so u can begin each experiment with a clean un thawed reproducible stock sample. doing this will help preventing damage to your main storage bank.
