Why growth in Tricine buffered medium is better to traditionally HEPES buffered medium for Scenedesmus at same pH? Even good buffers like MOPS and Gly glyc are not performing. What could be be possible explanation for the same.
Buffer solution having pKa value near to the target pH level is selected to maximize buffer capacity of the medium. I hope your organism grows near pH 7.8 and it is very sensitive too. All the buffer you tried have pka value below than TRICINE. I hope if you use HEPPS or EPPS buffer, which have pka value near to tricine will give similar result.
Hi Deepti, use tissue culture grade reagents. Believe me, it makes a lot of difference.

But these studies were carried out at pH-7.0. where the pKa values of HEPES, MOPS and GlyGly lie. Yet algae fail to grow in this medium but grows luxuriantly in Tricine at the same pH.
