We are unisng Dual luciferase assays for experiments trying to find interactions between proteins and the the recomendation for using the Dual luciferase assay from Promega is to use luminometers with injectors, which we don´t have (we have a multiplate luminometer). In the case fo not using injectors the recomendation is to use the Dual Glo luciferase but it s double the price fo the Dual Luciferase. So, I wonder if anyone has used the Dual Luciferase assay without injectors. Also has anyone use the Pierce™ Renilla-Firefly Luciferase Dual Assay Kit which is similar to the Dual luciferase assay from Promega.
The Dual Luciferase kit from Promega requires injectors because the signal half-life is about 1 minute. This can lead to a large amount of variation across a plate if done manually. I didn't see any details on the signal half-life of the Pierce kit, but I would assume that it also has a very short half-life.
I can attest to the usefulness of the Dual Glo assay from Promega if you have a plate reader without injectors. The cost might be more than the Dual Luciferase assay, but it is the only kit I'm aware of that works reliably in that situation.
